Traveled through there about 5 years ago while traveling from Germany to Milan. Stayed in Prague for 3/4 days. It's lost a lot of it's 'magic' that people use to love about it because it's a lot more main stream touristy now. Just my opinion though.

prague is nice. lots of touristy stuff like the bridge. the night club scene was disappointing but I only tried one. pretty dead actually. strip clubs are awesome.
go to vienna too via train. great food there.
unlike france, italy etc, the hotels are pretty nice and modern for about $150-200 / night.

I'm almost a regular to CZ, although I'm almost 1000 km away from Brno, CZ.
Have been to Prague several times, liked it pretty much, but Brno is the city I love. I think I have been there 5-6 times in the past 12 months.
Its just so beautiful and as being said before, Czech girls are smoking, especially in Brno.
If you decide to stay there, get a room at the Barcelo Brno Hotel.
